Protein Profiling of Orthosiphon stamineus Embryogenic Callus using Liquid Chromatography Coupled with Tandem-Mass Spectrometry (LC-MS/MS)

Orthosiphon stamineus is one of the most prominent medicinal herbs widely grown in Southeast Asia. Propagation O. using tissue culture technique helps to rapidly produce samples for research purposes, increase production secondary metabolites and considered as strategies plant genetic improvement. Despite vast information on pharmacological properties, protein profile this species currently underexplored. In present study, proteins expressed embryogenic callus developed from leaves Murashige Skoog medium supplemented with 1-naphthaleneacetic acid (NAA) 2,4-dichlorophenoxyacetic (2,4-D) were identified via Liquid chromatography- tandem mass spectrometry (LC-MS/MS). The presence was confirmed microscope observation Evan Blue assay. Using proteomic approach, we peptides that represent 22 localized different cell compartments involved various biological metabolic functions plants. This finding useful improve our understanding functions, thus their future applications could be further explored. Keywords: Embryogenic, Callus, Herb, stamineus, Proteins
